Can I restrip and reprobe a sequenced plate?

Yes. Restripping and reprobing a prepared plate is possible by following the stripping and rinse protocol below (NOTE: the recipeit is confidential and make sure to consult with FAS first before disclosing)

1. Stripping Buffer Formulation (Buffer 1) -
Prepare Buffer 1 using the following final concentrations (brought to final volume with nuclease-free water):

60% Formamide

0.2X SSC

0.1% Triton X-100

Nuclease-free water (remainder)

2. Stripping & Wash Protocol
Stripping Step: Incubate the sample with Buffer 1 for 10 minutes, repeated 3 times (3 × 10 min). (Note: There should still be stripping buffer remaining in the bottle after completing the run.)

Rinse Step: Rinse with PBST for 10 minutes, repeated twice (2 × 10 min) to clear residual stripping reagents before reprobing.
